I suggest you create a cover rather than a bag to be worn on the back. It is not easy to carry something that heavy on the back and that too..via stairs. I suggest you create a bag in a horizontal style and place the bottle as well in the horizontal way..like a duffle bag..and make a super comfortable cushion based handle or something comfortable
